Cytokines Series: Brain Derived Neurotrophic Factor -BDNF

In the brain, BDNF is expressed predominantly in the hippocampus, cortex, and synapses of the basal forebrain areas where it is vital to learning, memory, and higher thinking. BDNF is also expressed in muscles. BDNF selectively supports the survival of primary sensory neurons, retinal ganglia and the survival and differentiation of certain cholinergic and dopaminergic neurons. It also modulates neural plasticity and predisposition to behaviors. BDNF can recover re-innervation in the mature cerebellum thus it is applicable in trauma of the central nervous system (CNS). Both BDNF and NGF express neuroprotective effects on myelin, thus they are indicated in multiple sclerosis (MS). Exposure to stress and the stress hormone corticosterone has been shown to decrease the expression of BDNF which can lead to depression. On the other hand, the expression of BDNF is increased by voluntary exercise, caloric restriction, intellectual stimulation, curcumin, the neurotransmitter glutamate and various antidepressants drugs.BDNF is also a critical regulator of drug dependency since it causes cravings in opiate-dependent people; on the other hand BDNF has a role in maintaining long-term abstinence from alcohol in people with alcohol dependency.

Cytokines Series: Nerve Growth Factor – NGF

This video introduces the cytokine Nerve Growth Factor – NGF, an important nerve regenerative and repairing neurotrophine. NGF is synthesized in the hypothalamus, pituitary, thyroid gland, testis, and it is also produced by vascular smooth muscle cells and fibroblasts. The expression of NGF in specific neurons of the central nervous system (cortex, hippocampus) can be influenced positively by glutamate and negatively by GABA stimulating neuronal activity. NGF stimulates the growth and differentiation of B-cells and the growth of T-cells and also promotes the proliferation of mast cells. Vitamin D3 is potent inducer of NGF synthesis (a possible reason for vitamin D antidepressant action), while glucocorticoids are potent NGF inhibitors.

Cytokines Series: Insulin Like Growth Factor 1 – IGF 1

This is the thirteenth of a series of short videos on Low dose Medicine and in particular Cytokines. This video introduces the cytokine IGF 1 , the ‘anti age’ growth factor. Insulin-like growth factor 1 (IGF-1), is a protein similar in molecular structure to insulin mainly produced by the liver. Its production is stimulated by the growth hormone (GH) and can be retarded by under nutrition or malnutrition, growth hormone insensitivity, lack of growth hormone receptors and it is also reduced with age. In children, it stimulates bone growth and development of organs such as the heart, liver, and kidneys. In humans, the growth hormone (GH) made in the anterior pituitary gland stimulates the liver to produce insulin-like growth factor 1 (IGF-1) which then stimulates and has a growth-promoting effect on almost every cell in the body, especially kidney, muscles, cartilage, liver, nerves, skin, hematopoietic cells, and lungs.

Cytokines Series: Tumor Necrosis Factor – TNF α

This is the fourteenth of a series of short videos on Low Dose Medicine and in particular Cytokines. This video introduces the cytokine Tumor Necrosis Factor α , an important inflammatory cytokine. TNF-alpha has a wide spectrum of biological activities, generally together with IL-1 and IL-6, such as prostaglandin synthesis by hypothalamic cells, HPA (hypothalamic-pituitary-adrenal) axis stimulation by stimulating the release of corticotropin releasing hormone (CRH), appetite suppression, fever induction, hepatocytes stimulation for the production of acute phase proteins, hypoglycemia, insulin resistance and stimulation of phagocytosis on macrophages. High concentrations of TNF induce shock-like symptoms whereas the prolonged exposure to low concentrations of TNF can result in cachexia.

Cytokines Series: Granulocyte Colony Stimulating Factor – The immune system trigger

This is the eleventh of a series of short videos on Low dose Medicine and in particular Cytokines. This video introduces the cytokine Granulocyte colony-stimulating factor (G-CSF or GCSF). Granulocyte colony-stimulating factor is a glycoprotein which stimulates the the proliferation and differentiation of hematopoietic progenitor cells from the bone marrow into granulocytes (white blood cells) and neutrophils. G-CSF is produced by monocytes, macrophages, and neutrophils after cell activation and it is also produced by stromal cells, fibroblasts, and endothelial cells. G-CSF triggers the reactivity and raises the sensitivity of the immune system, and it enhances the antibody-dependent cell mediated cytotoxicity of granulocytes against tumor cells.

Cytokines Series: Interleukin 12 – The Allergy Support Cytokine

This is the tenth of a series of short videos on Low dose Medicine and in particular Cytokines. This video introduces the cytokine Interleukin 12. IL 12 is also known as a T cell-stimulating factor because it can stimulate the growth and function of T cells and especially the differentiation of naive T cells into Th1 cells (cell mediated immunity). IL-12 enhances the cytotoxic activity of NK cells and CD8+ cytotoxic T lymphocytes and also induces the synthesis of IFN-gamma, IL2, and TNF-alpha (TNF-α). IL-12 inhibits the synthesis of IgE induced by IL-4 and can therefore be useful in allergic diseases. IL 12 has anti-angiogenic (it can block the formation of new blood vessels), anti-tumoral and anti-metastatic activity, and because of this, it has been proposed by immunologists as a possible anti-cancer drug (although in pharmacological doses it can cause many side-effects). IL 12 is the most important cytokine with IL 2 and interferon gamma (IFN-gamma) in neoplastic pathologies (cancer support therapy) in general and especially melanomas and kidneys carcinoma.

Cytokines Series: Interleukin 10 – The Master Anti-inflammatory Cytokine

This is the ninth of a series of short videos on Low dose Medicine and in particular Cytokines. This video introduces the cytokine Interleukin 10. IL10 regulates the reactivity of the organism and inhibits the synthesis of most pro- inflammatory cytokines such as IL1, IL6, IL8, IL12, IL23 and tumor necrosis factor (TNF) and also the Th1 sub populations of T-cells such as IFN-gamma and IL2. IL 10 regulates cell cycles and prevents nitric oxides and oxidative stress, it suppresses metastasis and tumor invasion. Since it inhibits the release of IL 1 and TNF it is able to slow down cartilage erosion. IL-10 can stimulate the synthesis of IgE antibodies and acts synergically with IL 4 to inhibit cell mediated immunity. It also prevents insulin resistance and insulin sensitivity.

Cytokines Series: Interleukin 7 – The Lymphoid progenitor Cells Cytokine

This is the eighth of a series of short videos on Low dose Medicine and in particular Cytokines. This video introduces the cytokine Interleukin 7. IL 7 stimulates the hematopoietic stem cells into lymphoid progenitor cells and the proliferation of all cells in the lymphoid lineage such as B cells, T cells and NK cells and in particular it stimulates the antitumoral action of cytotoxic T lymphocytes. Since it acts as a lymphocytes growth factor it is of particular importance for B and T cell survival and development. IL7 is critical for immune homeostasis and it could also be beneficial in improving immune recovery after stem cell transplant.

Cytokines Series: Interleukin 4 – The anti -inflammatory cytokine

IL 4 is the main cytokine to induce differentiation of naive T helper cells into T helper 2 (Th2) cells which, upon activation, subsequently produce additional IL-4.IL 4 is important in generating protective immunity against all parasitic infections. It stimulates the proliferation of B lymphocytes and mast cells and induces a B-cell class switching to IgE antibodies.IL-4 directs bone marrow progenitor cells to differentiate into Th2 cytokine-producing eosinophils. It is antagonist to the production of Th1 cells, macrophages, IFN-gamma, and IL-12. It also inhibits cell activation of NK cells induced by IL2.IL4 is important in the treatment of inflammatory diseases and autoimmune diseases since it inhibits the production of inflammatory cytokines such as IL1, IL6 and TNF- alpha by monocytes and by T-cells.IL 4 also helps the immune system to deal with environmental and industrial toxins and plays a critical role in the higher functions of the brain, such as memory and learning.IL4 is involved in the pathogenesis of chronic lymphocytic leukemia disease by preventing both the death and the proliferation of the malignant B-cells.IL-4 is the key inducer of all allergic reactions.

Cytokines Series: Interleukin 2 – The ‘long life’ cytokine

This is the fifth of a series of short videos on Low dose Medicine and in particular Cytokines. This video introduces the cytokine Interleukin 2. IL 2 stimulates T and B lymphocytes and NK cells; it regulates the adaptive immune system and stimulates cell-mediated immune responses. IL 2 is a ‘master’ cytokine for the discrimination between self and non-self which is a hallmark of healthy immune system. In fact, IL-2 is necessary during T cell development in the thymus for the maturation of regulatory T cells. IL 2 is also pivotal for the growth, proliferation, and differentiation of T cells to become ‘effector’ T cells. IL 2 is able to facilitate antibody production and response, especially immunoglobulins produced by B cells. The production of IL 2 decreases with age (along with the hormone Melatonin and the growth factor IGF-1) which is one of the reasons why elderly people are more susceptible to infections and cancer.
